Is anybody else using the bioruptor (diagenode) to sonicate yeast (I'm using S.pombe) for ChIP on chip? I'm currently using 8 cycles of 30s ON 60s OFF on high setting to sonicate 2ml lysate (from 100ml cells at OD600 = 1) to get an average fragment size of around 400-500bp. I've done a time course up to 16 cycles (30s on 60s OFF) where I get ~ 200bp fragments. I was thinking about increasing the number of cycles to see whether there is a significant effect on resolution. I'd be interested to know if anybody else has tried different sonication conditions using the biorupter for yeast ChIP on chip and what conditions you find work best.
